  • 美國 Akoya Biosciences 公司開發之 CODEX（CO-Detection by indEXing）系統，為多重免疫組織影像分析領域的重要創新平台。此系統利用專屬的寡核苷酸條碼（Barcodes）標記於單株抗體上，可同時將多種標的抗體施加於同一張組織切片，實現一次性標記所有目標蛋白。其影像擷取過程採「循環式螢光顯示」原理，透過具螢光團的互補寡核苷酸序列（Reporters）依序分批作用，每輪可顯示三種標的物並完成影像擷取，接著移除螢光團後再進行下一輪反應。此循環可進行超過40輪，使得單一組織切片可同時觀察 40種以上蛋白標記，遠超傳統免疫螢光技術僅能同時觀察4–6種標記的限制。

    The CODEX (CO-Detection by indEXing) System developed by Akoya Biosciences (USA) is an innovative platform for highly multiplexed immunofluorescence imaging. It uses unique oligonucleotide barcodes conjugated to monoclonal antibodies, allowing simultaneous labeling of multiple protein targets on a single tissue section. The imaging process operates through cyclic fluorescence detection, where fluorescently tagged complementary oligonucleotides (reporters) bind to target barcodes in groups of three for imaging. After each cycle, the reporters are removed and replaced for the next round. This enables over 40 protein markers to be visualized on a single tissue section —far surpassing the 4–6 marker limit of conventional immunofluorescence.

    可自動完成染色、拍攝、去除螢光等流程，並自動重複循環進行，大幅減少人力並提升實驗效率。 / The fluidics system automates staining, imaging, and fluorophore removal in repeated cycles, reducing manual workload and improving throughput.

  • 多重影像分析軟體 | Multiplex Analysis Software (MAV)
    可執行影像漂移校正、背景扣除、細胞分割、聚類及空間網絡分析，生成可視化的組織空間圖。 / The MAV software performs image alignment, background subtraction, cell segmentation, clustering, and spatial network mapping to produce high-dimensional spatial visualizations.
  • 高多重標記能力 | High Multiplexing Capability
    可於同一組織切片上同時觀察超過 40 種以上標記蛋白質。 / Enables simultaneous visualization of more than 40 protein markers within a single tissue section.

  • 組織樣本相容性 | Sample Compatibility
    適用於冷凍切片及石蠟包埋組織切片（FFPE），經處理後仍可維持組織結構完整，並可再進行 H&E 染色。 / Compatible with both frozen and FFPE tissue sections. After processing, tissue integrity is preserved for subsequent H&E staining if needed.

申請流程 & 注意事項 / Application & Notes

使用儀器前，請務必詳細閱讀 CODEX實驗流程注意事項

  • 使用者需先與技術員討論實驗設計，完全了解流程並完成繳費後，方可預約儀器。 / Users must consult with a technician regarding experimental design and fully understand the workflow; reservation is allowed only after payment is completed.

  • 儀器採網路預約制，請依據抗體數量及檢體片數預約所需使用時數。 / Instrument reservation is conducted online; schedule usage hours according to the number of antibodies and tissue sections.

  • 每次預約須與前一位使用者完成上機後間隔至少 3 天。 / Reservations must be at least three days apart from the previous user’s session.

  • 組織切片及抗體標記需自行準備與操作，本組僅提供操作指導，不協助實驗操作。 / Tissue sections and antibody labeling must be prepared by the user; staff provide guidance only, no hands-on assistance.

  • 每片切片建議至少標記 6 支抗體，抗體數量建議為 3 的倍數。 / Each section should be labeled with at least six antibodies; antibody numbers are recommended in multiples of three.

  • 儀器全程由技術員操作，使用者不得擅自操作儀器。 / The instrument is operated entirely by technicians; users may not operate it themselves.

  • 儀器運作與資料處理期間，請勿隨意接觸儀器及電腦。 / Do not touch the instrument or computer during operation and data processing.

  • 本組僅提供 gating 後分析資料，後續數據分析需使用者自行完成。 / Only post-gating analysis data are provided; further data analysis must be performed by the user.

  • 研究資料須使用已格式化隨身硬碟，經技術員確認後方可存取。資料保存期限為 6 個月，逾期資料將依雙方同意後刪除。 / Research data must be saved on a pre-formatted external drive; access is allowed after technician confirmation. Data are stored for six months and deleted after mutual agreement.

  • 儀器故障時，請立即通知管理人員，勿自行拆裝或調整參數。 / Report any instrument malfunction immediately; do not disassemble or adjust parameters yourself.

  • 因個人操作不當造成儀器損壞，維修費用由使用者所屬實驗室負擔。 / Any damage caused by improper user operation will be charged to the user’s laboratory.
