儀器簡介 / Instrument Introduction

  • FV1000 為高解析度共軛焦雷射掃描顯微鏡，搭配倒立式或正置式顯微鏡，可進行活細胞或固定樣品觀察。採用多波長雷射與高靈敏偵測器，可同時拍攝多螢光通道，並搭配高速掃描與多點掃描技術，以取得高品質影像。可進行三維重建 (3D reconstruction)、光漂白 (FRAP)、光轉換 (Photoconversion) 與活體動態觀察。

  • The FV1000 is a high-resolution confocal laser scanning microscope, compatible with inverted or upright microscopes for live-cell and fixed specimen imaging. It uses multiple wavelength lasers and high-sensitivity detectors for simultaneous multi-channel fluorescence imaging. High-speed and multi-point scanning provide high-quality images, suitable for 3D reconstruction, photobleaching (FRAP), photoconversion, and live-cell dynamic studies.

儀器規格 / Instrument Specifications

  • 顯微鏡系統：OLYMPUS IX-81 倒立式顯微鏡，具自動對焦與 Z 軸漂移補償。/ Olympus IX-81 motorized inverted microscope with autofocus and Z-drift compensation.

  • 雷射波長：405nm、440nm、488nm、515nm(不支援)、559nm、635nm；另配 SIM 405nm 雷射用於光刺激。 / Laser wavelengths: 405, 440, 488, 515 (not supported), 559, 635 nm; plus SIM 405nm laser for photo-stimulation.

  • 物鏡倍率：10X、20X、40X(Oil)、60X(Oil)、100X(Oil)，含 DIC。 / Objectives: 10X, 20X, 40X(Oil), 60X(Oil), 100X(Oil) with DIC.

  • 三頻道同時掃描，配有 DIC，若含 DIC 可同時四頻道掃描。 / Three fluorescence channels + DIC; four-channel scanning available with DIC.

  • 馬達化工作台與鼻輪，可精準選取位置並進行馬賽克掃描。 / Motorized stage and nosepiece for precise position selection and mosaic imaging.

  • 馬達化孵育室支援 37℃、5% CO2、90% 濕度，可進行長時間活細胞觀察。 / Stage-top incubator for 37°C, 5% CO2, 90% humidity; suitable for long-term live-cell imaging.

  • 高靈敏度偵測器，支援微弱訊號與超解析應用。 / High-sensitivity detectors for weak signal and super-resolution applications.

申請流程 & 注意事項 / Application & Notes

  • 自行上機 / Self-Operation

    1. 使用者須通過本中心兩階段認證考試合格後，方可預約並自行操作，並負責儀器安全。 / Users must pass the two-stage certification exam conducted by the center before booking and operating the instrument independently. Users are responsible for instrument safety.

    2. 預約前請至中心網站登記並詳閱使用規則。 / Please book the instrument via the center website and read the usage rules carefully.

    3. 若有國科會計畫者，請至國科會基礎研究核心設施預約服務管理系統預約儀器 / For MOST-funded projects, users must also book the instrument via the MOST Core Facilities Reservation System.

    4. 使用結束後，操作管理人員會依使用時段開立繳費單，使用者持繳費單至出納組繳費，再將繳費單繳回操作管理人員。 / After use, staff will issue a payment slip according to usage hours. Users pay at the university cashier and return the slip to the staff.

  • 委託上機 / Staff-Assisted Operation

    1. 技術員操作儀器，使用者在旁說明實驗目的及影像需求。 / Staff operates the instrument while users explain experiment purpose and imaging requirements.

    2. 請事先與操作管理人員預約使用時段。 / Users must book a session with staff in advance.

    3. 若有國科會計畫者，請至國科會基礎研究核心設施預約服務管理系統預約儀器 / For MOST-funded projects, users must also book the instrument via the MOST Core Facilities Reservation System.

    4. 下載填寫繳費單，委託完成後交由操作人員，依使用時數統計收費，再持繳費單至出納組繳費，完成後將繳費單交回操作管理人員。 / Download and fill in the payment slip. After staff-assisted use, submit it to the staff; pay at the cashier based on usage hours and return the slip.

    • 常規認證 / Regular Certification

      • 條件 / Requirement: 需自行操作者 / For self-operation users

      • 收費 / Fee: 免費 / Free

      • 認證流程 / Procedure:

        1. 參與教育訓練課程並通過筆試。 / Attend the training course and pass the written test.

        2. 課程含實機操作說明，後續通過實機操作考試。 / Course includes hands-on training; pass practical exam afterward.

        3. 通過後半年內完成第一次預約上機，由種籽師資陪同確認操作無誤，即可自行預約。 / Within six months, complete first supervised session; upon approval, user can book independently. 

    • 急件認證 / Urgent Certification

      • 條件 / Requirement: 急需自行操作者 / For urgent need of self-operation

      • 收費 / Fee: 教學認證 3 小時 + 第一次陪同上機 3 小時，依委託上機費用計算 (NT$2500/3小時)。 / Training + first supervised session, charged at NT$2500/3 hours.

      • 認證流程 / Procedure:

        1. 線上觀看課程影片，並預約筆試。 / Watch online course and schedule written test.

        2. 通過後與技術人員預約現場教學認證 (含實機操作練習及考試)。 / Schedule on-site training and practical exam with staff.

        3. 一個月內完成第一次上機，由技術人員陪同，確認操作無誤後，即可自行預約操作。 / Complete first supervised session within one month; after approval, user can book independently.

      附註 / Note: 若需額外陪同教學時數，每小時收費 NT$1000。 / Additional supervised training hours are charged NT$1000/hour.
